Urgent WordPress Plugin Fix

Two of my WordPress installs have suddenly lost most of their functionality because the plugins refuse to activate. No new plugins were installed and nothing was recently updated, so I need someone who can jump in right away, trace the underlying cause, and get every required plugin running again without taking the sites offline any longer than absolutely necessary.
