Statistical advice for which test to use

Hi

I have 3 questions about what's the ideal test to run for a couple of my study designs. I would appreciate any advice!

First question:
I have:

• One within-subjects factor: time (5 levels)

• 2 between-subjects factors: group (2 levels) and treatment (2 levels)

• And one dependent variable which is the levels of a biomarker in the blood.

I would like to see if the change in the biomarker levels significantly differ between the groups in response to the treatment over time.

Is it okay if I do a three-way(2x2x5) mixed ANOVA on this? A colleague told me that it is completely wrong to use an ANOVA and that I should do a linear mixed model instead.
.......

2nd question: I have another very similar study but with only one in-between groups factor. Is it fine if I use a two-way mixed anova?

.....

3rd question: If someone is familiar with graphpad prism can let me know if this test is suitable as an alternative for a two-way mixed anova in case of missing values. I also need info about it. Is it a form of a linear mixed model? https://www.graphpad.com/guides/prism/latest/statistics/stat_anova-approach-vs_-mixed-model.htm

Thanks in advance!
