2D segmentation in gray scale

I would like to create an interface that segments a particular area in gray scale values and gives the % in mm2. NOK & OK results.
Segmented area >40% of total area= NOK.
Segmented area <40% of total area= OK.
Red line to be segmented and blue line total area (this will be a static value).
