Plot a MapQuest Map in Real Time

I currently have a drag and drop list of clients (for a route) that the order can be changed and saved. This was done using jQuery and PHP.
I would like to have a MapQuest map beside this sortable list that plots all the destinations as numbers and shows the most efficient route between each client. As the sortable list is changed the map points and routes are also changed in real-time.
