Medical Image Classification Program

I need a program that can analyse SGRT body reference images and tell me how much darkness percentage in the image. I have attached a sample image. The program should be able to perform 3 D and 2D image analysis effectively on medical images. Ideal candidates for this project would be those with experience in developing image classification software like phython and MATLAB particularly for medical images.
