Classify Orphan Hydrolase Enzymes

Budget: £250 – £750 GBP

I am after unclassified — largely orphan — protein sequences and I need an enzymologist–bioinformatician to determine which of them truly belong to the hydrolase family. You will dive into the raw sequence data, interrogate it against UniProt, Pfam and InterPro, and support your calls with BLAST, EC-BLAST or comparable alignment and motif-search approaches.

The exact objectives (e.g., depth of annotation, EC number resolution, structural modelling) will be finalised with you once we start, but the core task is to produce a defensible list of candidate hydrolases together with the evidence behind every decision. Your enzymology background, familiarity Bioinformatics and functional annotation, and your ability to write up clear, reproducible methodology are essential.

Deliverables
• A detailed report and spreadsheet or database export listing every sequence examined, predicted activity, proposed EC number (where possible) and confidence level
• A concise methods document detailing search parameters, cut-offs, and reasoning
• Recommendations for follow-up validation (wet-lab or in-silico) where confidence is low
